Crystal structures of a nicotine MATE transporter provide insight into its mechanism of substrate transport

A transporter of the multidrug and toxic compound extrusion (MATE) family, Nicotiana tabacum MATE2 (NtMATE2), is located in vacuole membrane tobacco plant root involved transportation nicotine, a secondary or specialized metabolic Solanaceae. Here, we report crystal structures NtMATE2 its outward‐facing forms. The overall structure has bilobate V‐shape with pseudo‐symmetrical assembly N‐ C‐lobes. In one structure, C‐lobe cavity interacts an unidentified molecule that may partially mimic substrate. addition, NtMATE2‐specific conformational transitions imply unprecedented movement transmembrane α‐helix 7 related to release substrate into vacuolar lumen.
